Abstract
Sea grapes (Caulerpa racemosa) is a type of seaweed (macroalgae) that grows in Indonesia and is included in the chlоrорhуcеае class due to its green thаllus. These green macroalgae not only contain chlorophyll pigments but also carotenoids, which are photosynthetic pigments. This study aimed to isolate β-carotene from the pigment extract of Caulerpa racemosa by identifying the pigment using thin layer chromatography (TLC), then separating and purifying by column chromatography, and then characterized by UV-Vis spectrophotometry. The final stage of this study was to test the antimicrobial activity of β-carotene isolates against the test microbes Propionibacterium acnes, Staphylococcus aureus and Candida albicans. From this study, the pigment β-carotene was obtained with a concentration of 56.95 mg/L. Furthermore, β-carotene provided antimicrobial activity against Propionibacterium acne (15.65 mm), Staphylococcus aureus (17.1 mm), and Candida albicans (16.21 mm). The final results of this study showed the value of the minimum inhibitory concentration (MIC) of carotenoid pigments against Propionibacterium acne at a concentration of 50% (2.18 mm), against Staphylococcus aureus at a concentration of 50% (3.1 mm), and against the fungus Candida albicans at a concentration of 25% (2.98 mm).
